Dewey Mccafferty’s Expertise A central focus of the McCafferty laboratory is on developing chemical biology tools and conducting structural and mechanistic biochemistry to explore the molecular level mechanism, structure, and selectivity of enzymes and enzyme-containing multiprotein complexes involved in epigenetic modification of chromatin, in bacterial virulence and metabolism, and in oxidoreductase biological electron transfer. An additional mission is to connect these molecular level events to biological phenomena, to exploit critical enzyme functions in the design and evaluation of novel therapeutics for cancer, inflammation, and as antimicrobials, and understand the molecular underpinnings of enzyme dysfunction and its relationship to disease pathobiology. Prof. McCafferty has trained formally in organic and peptide chemistry. He was the Burroughs Wellcome Fellow in Synthetic Organic Chemistry at UNC and the Judith Graham Pool Postdoctoral Fellow at Harvard Medical School, working on the vitamin K-dependent carboxylase and VanX peptidase involved in vancomycin resistance. He spent a decade on the faculty at the University of Pennsylvania in the Biochemistry and Biophysics Department, where at that time he was selected for the Eli Lilly Award in Biological Chemistry, sponsored by the American Chemical Society. After moving to Duke University in 2006, Prof. McCafferty has developed new research programs in histone demethylation, chlamydial pathogenesis, mechanistic enzymology of natural product biosynthesis and natural product mechanism of action. Prof. McCafferty has 16+ years experience in mechanistic enzymology, epigenetics, construction of synthetic chromatin templates, synthetic organic and peptide chemistry, protein design, protein biophysics and most recently macromolecular protein crystallography. He currently is the Chair-elect for the American Chemical Society Division of Biological Chemistry.
