Maleana Khoury’s Expertise I am a self-driven and highly motivated plant molecular/developmental biologist. My areas of training and expertise have centered around cell-cell and hormone signaling, genetics and genomics, and (broadly) plant molecular biology. I have extensive skill in a broad range of molecular techniques. I have performed RNA extraction using both phenol-chloroform and solid-phase extraction methods. I have employed Primer3Plus, PrimerQuest (IDT), and Integrated Genome Browser to design primers for PCR and qPCR. I have performed two-step RT-qPCR using QuantStudio3 equipment and software and performed data analysis using both standard curve and delta delta Ct methods. I have employed both Gibson Assembly and restriction enzyme digest for molecular cloning and have used both E. coli and S. cerevisiae recombinant expression systems. I have used the Agrobacterium-mediated transformation method to clone reporter and over-expression constructs into Arabidopsis thaliana and used the selectable herbicidal marker glufosinate ammonium to identify transformed lines. I have performed imaging of both live and fixed tissue and cells using an epifluorescence microscope (Echo Revolve) and laser scanning confocal microscopes (Olympus and Zeiss) in combination with fluorescent endogenous reporters and dyes and immunofluorescence staining techniques. I have conducted thin tissue sectioning using both vibratome and cryostat technology. I have executed pharmacological inhibitor studies using DAPT (N- [N- (3, 5-difluorophenacetyl)-l-alanyl]-s-phenylglycinet-butyl ester) and quantified cell proliferation using EdU. I have performed immunoblot analysis using dry transfer methods and run samples on SDS-PAGE gels. I have maintained cell cultures of immortal cancer cell lines and used Comet Assay to quantify DNA damage. I have proficiency in R, RStudio, Excel, and GraphPad for data analysis and visualization. I have mentored both undergraduate and graduate students in both authentic research and classroom settings and have delivered guest lectures in advanced undergraduate-level courses. I have an unmatched work ethic and enthusiasm in the workplace and work well as part of a collaborative team. On the other hand, I am highly self-motivated and self-taught and function well as an independent scientist. I have a strong ability to troubleshoot and problem solve in addition to conducting rigorous and reproducible research. I have contributed to the generation of multiple peer-reviewed publications in established journals during my undergraduate and graduate years.
