Robert Mclaren’s Expertise Have spent the past 25 years at Promega working with the Genetic Identity group. This group is involved in developing solutions for DNA typing for the forensic and paternity DNA testing communities. Currently I am associate director of the Genetic Identity R&D group. My experience started with the technical support and training before moving into R&D in 2007, and has involved development of automated methods for various Promega chemistries, support and training on expert system softwares. Have spent the past 16 years in R&D developing various and brand new multiplex STR systems (e.g. original PowerPlex ESX and ESI Systems, and PowerPlex 21 System), including rapid cycling chemistries (e.g. PowerPlex ESI Fast and ESX Fast Systems), and a suite of reagents (e.g. SwabSolution) that enable direct amplification from a variety of single source samples like buccal swabs and nonFTA cards. Also led the group of R&D scientists that developed the PowerQuant System for the quantification of total human and male genomic DNA. This new quantification system for human DNA also includes a target that allows for the evaluation of sample integrity prior to performing STR amplification. More recently, was the technical lead on the 2020 launch of the Spectrum Compact capillary electrophoresis (CE) system in collaboration with Hitachi High-Technologies. In July 2020, became responsible for the Genetic Identity R&D group, being promoted to Associate Director in September 2022. In that time, the Genetic Identity group have seen the launch of the very first commercially available 8 color STR System (PowerPlex 35GY) for use on the Spectrum CE System (first commercially available 8 color CE system). Specialties: Developing new multiplex PCR chemistries for analysis of STRs. Developing solutions for direct amplification with STRs and qPCR Developing gPCR assays Developing rapid cycling PCR chemistries Automating chemistries on liquid handling platforms such as Beckman Coulter's Biomek 3000 liquid handling system. Providing training, support and feedback on software for the analysis of STR data. Root cause analysis of technical issues affecting PCR and capillary electrophoresis.
