Usha Ghosh’s Expertise (I) Lead a team involved in design and synthesis of NCEs based on mechanism of action for cardiovascular, anticancer and anti-inflammatory indications. NCEs based on imidazopyridine scaffold were synthesized and an orally active tumor necrosis factor-α-inhibitor was developed as a drug candidate for rheumatoid arthritis and inflamatory bowel disease. NCEs having 3-((5-phenyl-1,3,4-oxadiazol-2-yl)thio)benzo[b]thiophene 1,1-dioxide and 11H-dibenzo[b,f][1,4]oxathiepine derivatives were also developed for the same. Substituted 3-(phenylthiomethyl)-4h-chromen-4-ones based NCEs were designed, synthesized as potent inhibitors of inflammatory cytokines for the colitis treatment. Design and synthesis of isocytosine based NCEs were achieved for xanthine oxidase inhibitors by lead optimization. Designed and synthesized a large number of 5- and 6 membered diazenic heterocyclic molecules as non-steroidal inhibitors for estrogenic receptors involved in breast cancer. (II) Worked for process R&D with kilo lab/ pilot plant/ production scale. Process developed for Vitamin-A, managed troubleshooting in each stage and achieved the production of pure Vitamin-A palmitate. Few APIs synthesised by enzymatic resolution, process development for Chlomipramine and Leuprolide. (III) Developed a new approach to peptides based on redox strategy. The method could be used for fragment coupling and cyclic peptide synthesis. A number of Enkephalins/ analogs and cyclic peptides including Antamanide have been made. Peptides with free C-terminal residues and peptides with unnatural amino acids were made. In organosilicon chemistry, developed method for acylsilanes. Achieved the advanced intermediates for (+)- and (-)- nonactic acids. I revealed for the first time, an extraordinary reaction of phenyldimethylsilyllithium with N,N-disubstituted amides wherein enediamines are formed. Ene diamines are intermediate in organic synthesis especially to make α-amino ketones.
